When a model of replicating molecules was created, it was found that, for effective storage of information, macromolecules on prebiotic Earth could not exceed a certain threshold length. This problem is known as the error threshold problem. It arises because replication is an imperfect process, and during each replication event, there is a risk of incorporating errors into a new sequence, leading to the creation of a quasispecies. In a system that is deprived of high-fidelity replicases and error-correction mechanisms, mutations occur with a high probability. As a consequence, the information stored in a sequence can be lost due to the rapid accumulation of errors, a so-called error catastrophe. Moreover, it was shown that the genome size of any organism is roughly equal to the inverse of mutation rate per site per replication. Therefore, a high mutation rate imposes a serious limitation on the length of the genome. To overcome this problem, a more specialized replication machinery that is able to copy genetic information with higher fidelity is needed. Manfred Eigen suggested that proteins are necessary to accomplish this task. However, to encode a system as complex as a protein, longer nucleotide sequences are needed, which increases the probability of a mutation even more and requires even more complex replication machinery. === Hypertension === As with other angiotensin II receptor antagonists, candesartan is indicated for the treatment of hypertension. Candesartan has an additive antihypertensive effect when combined with a diuretic, such as chlorthalidone. It is available in a fixed-combination formulation with a low dose of the thiazide diuretic hydrochlorothiazide. Candesartan/hydrochlorothiazide combination preparations are marketed under various trade names including Atacand Plus, Hytacand, Blopress Plus, Advantec and Ratacand Plus.

=== SSAP === The SSAP (Sequential Structure Alignment Program) method uses double dynamic programming to produce a structural alignment based on atom-to-atom vectors in structure space. Instead of the alpha carbons typically used in structural alignment, SSAP constructs its vectors from the beta carbons for all residues except glycine, a method which thus takes into account the rotameric state of each residue as well as its location along the backbone. SSAP works by first constructing a series of inter-residue distance vectors between each residue and its nearest non-contiguous neighbors on each protein. A series of matrices are then constructed containing the vector differences between neighbors for each pair of residues for which vectors were constructed. Dynamic programming applied to each resulting matrix determines a series of optimal local alignments which are then summed into a "summary" matrix to which dynamic programming is applied again to determine the overall structural alignment. SSAP originally produced only pairwise alignments but has since been extended to multiple alignments as well. It has been applied in an all-to-all fashion to produce a hierarchical fold classification scheme known as CATH (Class, Architecture, Topology, Homology), which has been used to construct the CATH Protein Structure Classification database.

==== Narcolepsy ==== An increased number of narcolepsy (a chronic neurological disorder) cases in children and adolescents was observed in Scandinavian and other European countries after vaccinations to address the H1N1 "swine flu" pandemic in 2009. Narcolepsy has previously been associated with HLA-subtype DQB1*602, which has led to the prediction that it is an autoimmune process. After a series of epidemiological investigations, researchers found that the higher incidence correlated with the use of AS03-adjuvanted influenza vaccine (Pandemrix). Those vaccinated with Pandemrix have almost a twelve-times higher risk of developing the disease. It only affected people who have the HLA-DQB1*0602 variant of the HLA-DQB1 gene, also known as DR2+. 0.02% of *0602 carriers who took the vaccine developed nacrolepsy, so while carrying *0602 and receiving the vaccine are necessary conditions, they are not sufficient. The missing conditions could be some other gene variants. There are three components in AS03: squalene, DL-α-tocopherol (vitamin E), and polysorbate 80. Squalene and polysorbate 80 both has long histoies of use in other vaccines and will not explain the uniqueness of the Pandemrix reaction, so vitamin E is the main suspect. The amount of vitamin E is no more than a day's normal dietary intake. Vitamin E increases hypocretin-specific fragments that bind to HLA-DQB1*0602 in cell culture experiments, lending credence to this theory. (A definitive proof would be a randomized controlled trial, but a "how many narcoleptics did we make" study would not pass ethical review.)
Other clinicians described a cluster of near‑fatal collapses between March and June 2016, including the deaths of two triplets on consecutive days. The prosecution alleged that two infants, Baby F and Baby L, had been deliberately poisoned with insulin, citing blood test results identified during the police investigation. Their lead expert witness described this as key evidence, though their interpretation has since been questioned by some specialists. The prosecution also argued that Baby M, the twin of Baby L, collapsed after air was injected into his bloodstream. Letby had volunteered for the shift on which Baby L collapsed, despite not being scheduled for night duty. Several consultants told the court they had previously raised concerns about Letby but were discouraged by hospital managers. One doctor recalled Letby making a remark shortly before an infant's death that he interpreted as inappropriate. The prosecution also presented a shift‑pattern chart showing Letby on duty for 25 incidents, supported by swipe‑card data. The chart has since been criticised by some statisticians as misleading. After Letby was removed from clinical duties, the unit's admission criteria were tightened to stop receiving the sickest babies, and more consultants were hired; the unexpected deaths ceased. The prosecution further alleged that Letby altered times on patient records; she denied this, saying any discrepancies were errors.
