This is a working overview of ACTH(4-10), written for readers who want more than a one-paragraph summary but less than a textbook.
This page was last updated on 2025-09-21 and is reviewed periodically as new material appears.
Development is attributed to researchers at the Institute of Molecular Genetics in Moscow during the early 1980s, building on earlier Soviet work with ACTH fragments. Russian regulatory approval followed for intranasal use, and the compound has remained commercially available there for decades. Most published human data originate from Russian and, later, some Eastern European clinical reports, which are not always accessible in English translation. Outside that region the material is generally handled as a research chemical rather than a licensed medicine.
Regulatory status differs sharply between jurisdictions. In Russia the peptide is registered as a prescription nasal preparation, while agencies such as the United States Food and Drug Administration have not approved it for any indication. Products sold elsewhere are typically labeled for laboratory research only, and such labels shift responsibility for safe handling to the purchaser. Because the same name covers pharmaceutical-grade nasal drops and bulk research powder, identity and purity documentation becomes the main practical concern when comparing sources.
Semax is a synthetic seven-amino-acid peptide whose sequence extends the ACTH(4-10) fragment with a C-terminal proline-glycine-proline tripeptide. The commonly cited sequence is Met-Glu-His-Phe-Pro-Gly-Pro, giving a molecular formula near C37H51N9O10S and a molecular weight close to 813.9 g/mol. It belongs to the broader class of synthetic ACTH fragments studied for central nervous system effects rather than for adrenal steroid stimulation. In practice the material appears as a lyophilized white powder for laboratory work or as a dilute saline solution in clinical settings.
Pharmacological accounts link semax to melanocortin signalling and to modulation of neurotrophic factor expression, particularly brain-derived neurotrophic factor and nerve growth factor. Much of this evidence comes from rodent studies using intranasal delivery, a route chosen because it allows peptides to reach the central nervous system with limited systemic exposure. Whether the same mechanisms operate in humans at comparable magnitude remains an open question. The precise receptor or receptors responsible for the reported behavioural and neuroprotective effects have not been conclusively identified.
Clinical reports describe use in ischaemic stroke, transient ischaemic attack, optic nerve conditions, and cognitive complaints, but most of these studies are small and were conducted in a single region. Systematic reviews have generally described the evidence base as limited in size and variable in methodological quality. Randomised controlled data suitable for international regulatory assessment are scarce. As a result, major treatment guidelines outside Russia do not include the peptide, and interest in it remains largely research-driven rather than routine clinical.
Semax is a synthetic heptapeptide with the sequence Met-Glu-His-Phe-Pro-Gly-Pro. It is described in the literature as an analogue of the ACTH(4–10) fragment, a short N-terminal portion of adrenocorticotropic hormone that retains some neurotropic activity without the full hormonal effects of the parent peptide. The molecule carries a methionine residue at the N-terminus and two proline residues near the C-terminus, features that shape both its interactions with receptor systems and its chemical stability. The free peptide corresponds to the formula C37H51N9O10S and a molecular mass near 813.9 Da.
| Property | Value | Notes |
|---|---|---|
| Chemical class | Synthetic heptapeptide (ACTH fragment analog) | Not a steroid; does not belong to the melanocortin agonist drugs by marketing category |
| Molecular formula | C37H51N9O10S (commonly cited) | Reported values vary slightly with salt and counter-ion content |
| Appearance | White to off-white lyophilized powder | A single batch may appear as a loose cake or fluffy solid |
| Solubility | Freely soluble in water and aqueous buffer | Stock solutions are usually prepared in sterile water or saline |
| Typical storage | −20 °C or below, desiccated and protected from light | Seal opened vials promptly to limit moisture uptake |
Laboratory descriptions of the material are consistent across suppliers. It appears as a white to off-white lyophilized powder that dissolves readily in water and in polar organic solvents such as dimethyl sulfoxide. Aqueous solutions are clear and colorless at low concentrations. Because the peptide contains methionine, oxidation at the sulfur atom is a recognized degradation pathway, and handling notes usually call for protected, desiccated storage. Reported purity for research-grade lots is generally above 95 percent as measured by reversed-phase high-performance liquid chromatography.
The compound is registered in Russia as a pharmaceutical product, most commonly formulated as a nasal solution, and has been used in that setting since the 1990s. Outside that jurisdiction it is generally handled as a research chemical rather than an approved medicine. Regulatory status therefore differs sharply between countries, and material sold internationally may not correspond to the Russian pharmaceutical formulation. Documentation with commercial samples is typically limited to a certificate of analysis covering purity and identity, not clinical status or local legal classification.
Semax is a synthetic heptapeptide whose sequence is Met-Glu-His-Phe-Pro-Gly-Pro. It was developed as a fragment analog of adrenocorticotropic hormone, modeled specifically on the ACTH(4-10) region. The first four residues reproduce that fragment, while a Pro-Gly-Pro tripeptide is appended at the C-terminus. Work on the compound originated in Russia, where it entered clinical use as an intranasal preparation. Its sequence places it among short regulatory peptides studied for effects on the central nervous system rather than on the adrenal axis.
The C-terminal Pro-Gly-Pro extension is not incidental. Proline-rich tails are known to resist several common peptidases, and the published literature attributes the longer half-life of Semax, relative to unmodified ACTH fragments, to this feature. The modification also removes the melanocyte-stimulating and corticosteroidogenic activity that characterizes longer ACTH-derived sequences. Because the molecule is small and hydrophilic, it is typically formulated as an aqueous solution for intranasal or parenteral delivery. Acetylation or amidation at the termini appears in closely related research peptides and shifts the mass by a fixed increment.
== Structure == GRF (1-29), also known as sermorelin (Tyr-Ala-Asp-Ala-Ile-Phe-Thr-Asn-Ser-Tyr-Arg-Lys-Val-Leu-Gly-Gln-Leu-Ser-Ala-Arg-Lys-Leu-Leu-Gln-Asp-Ile-Met-Ser-Arg-NH2), the biologically-active portion of the 44 amino acid GHRH. Half-life "less than 10 minutes", perhaps as low as 5 minutes. Mod GRF (1-29) replacement of the 2nd, 8th, 15th, and 27th amino acids of GRF (1-29) yields modified GRF(1-29) (Tyr-D-Ala-Asp-Ala-Ile-Phe-Thr-Gln-Ser-Tyr-Arg-Lys-Val-Leu-Ala-Gln-Leu-Ser-Ala-Arg-Lys-Leu-Leu-Gln-Asp-Ile-Leu-Ser-Arg-NH2). Half-life at least 30 minutes.

=== Metabolic detoxification === An animal's metabolism can produce harmful substances which it can then make less toxic through reduction, oxidation (collectively known as redox reactions), conjugation and excretion of molecules from cells or tissues. This is called xenobiotic metabolism. Enzymes that are important in detoxification metabolism include cytochrome P450 oxidases, UDP-glucuronosyltransferases, and glutathione S-transferases. These processes are particularly well-studied as part of drug metabolism, as they influence the pharmacokinetics of a drug in the body.
Archaeological evidence from sites in the United States, Mexico, and Peru indicates that mescaline-containing cacti have been used for over 6,000 years. Europeans recorded use of peyote in Native American religious ceremonies upon early contact with the Huichol people in Mexico. Other mescaline-containing cacti such as the San Pedro cactus have a long history of use in South America, from Peru to Ecuador. While religious and ceremonial peyote use was widespread in the Aztec Empire and northern Mexico at the time of the Spanish conquest, religious persecution confined it to areas near the Pacific coast and up to southwest Texas. However, by 1880, peyote use began to spread north of South-Central America with "a new kind of peyote ceremony" inaugurated by the Kiowa and Comanche people. These religious practices, incorporated legally in the United States in 1920 as the Native American Church, have since spread as far as Saskatchewan, Canada.

=== EPMA (Electron Plate Analysis) === EPMA determines elemental composition by bombarding a sample with an electron beam, causing it to emit characteristic X-rays. These X-rays are detected and analysed to identify elements. With high precision, EPMA is widely used in materials science, geology, and other fields. It helps understand material properties and investigate atomic-level details. By comparing detected X-rays with known standards, EPMA quantifies elements present in the sample.
Dry ice is produced industrially through the compression and cooling of carbon dioxide. The most common industrial method of manufacturing dry ice starts with a gas having a high concentration of carbon dioxide. Such gases can be a byproduct of another process, such as producing ammonia from nitrogen and natural gas, oil refinery activities or large-scale fermentation. The carbon dioxide-rich gas is then pressurized and refrigerated until it liquefies. Next, the pressure is reduced. When this occurs some liquid carbon dioxide vaporizes, causing a rapid lowering of temperature of the remaining liquid. As a result, the extreme cold causes the liquid to solidify into a snow-like consistency. Finally, the snow-like solid carbon dioxide is compressed into small pellets or larger blocks of dry ice. Dry ice is typically produced in three standard forms: large blocks, small (1⁄2 or 5⁄8 in [13 or 16 mm] diameter) cylindrical pellets and tiny (1⁄8 inch [3.2 mm] diameter) cylindrical, high surface to volume pellets that float on oil or water and do not stick to skin because of their high radii of curvature. Tiny dry ice pellets are used primarily for dry ice blasting, quick freezing, fire fighting, oil solidifying and have been found to be safe for experimentation by middle school students wearing appropriate personal protective equipment such as gloves and safety glasses. A standard block weighing approximately 30 kg (66 lb) covered in a taped paper wrapping is most common.
=== 1950–60s === After the war, some of the inventions found new applications. The first transistor radio was released in 1954, and its manufacturing process made use of vacuum pumps. The first integrated circuit or "chip" came to the market at the end of the 1950s, and vacuum pumps again played an important role in their production. These early inventions were Edwards' first steps into the electronics and semiconductors industry, which is to this day the company's biggest market. Chips became increasingly small throughout the decades, increasing the importance of vacuum purity.
=== Opposition to the Peace Preservation Act === In 1878, the Cape Parliament had passed the Peace Preservation Act, which allowed for the confiscation of the firearms of the African population in exchange for a monetary compensation. Sprigg decided that its implementation should extend to the Basuto, after witnessing 7,000 Basuto cavalrymen perform maneuvers during the course of Moorosi's uprising. This was announced during a pitso (formal assembly) attended by some 6,000 to 10,000 Basuto. Soon afterwards he also declared that the Quthing region would be confiscated by the Cape for white settlement. At the time almost half of all Basuto men owned a firearm. Many had worked in railway construction and the diamond mines in Griqualand West with the express purpose of purchasing modern breech-loading and smoothbore rifles. As a result, the Basuto became the best-armed tribe in southern Africa. For the Basuto gun confiscation was unacceptable, not only due to their high value, but also due to the necessity to defend their land and cattle in an environment where there was no guarantee of protection from the colonial authorities. For the Basuto, guns were a symbol of manhood, and to be disarmed was seen as being reduced to the status of a child. In April 1879, the Cape Colony doubled the hut tax in Basutoland to one £ per hut. In June 1880, Letsie I dispatched a three-man delegation to the Cape Parliament as it was deliberating the annexation of Quthing and the question of Basuto disarmament.

The new glass was taller and narrower than the previous one and featured a bevel design. The new glasses were planned to gradually replace the old ones. A practice known as "splitting the G" has been an entry on the Urban Dictionary website since 2018. This drinking game sees the drinker attempt to bring the liquid level of the Guinness to sit at a certain level in relation to the glass' Guinness branding. Variants of the practice see the required liquid level as either: between the letter 'G', on the horizontal line of the 'G', or between the 'G' and the harp logo.
An immigration judge rules that Trump can deport the Palestinian-born leader of Wisconsin's largest mosque—an outspoken advocate of Palestinian rights—who does not have a criminal record in the US, where he has lived since 1993 on national security grounds, citing his imprisonment in Israel in 1988 after being convicted of throwing stones at Israeli military personnel in the West Bank.
=== Medicine === Radioactive medical waste tends to contain beta particle and gamma ray emitters. It can be divided into two main classes. In diagnostic nuclear medicine a number of short-lived gamma emitters such as technetium-99m are used. Many of these can be disposed of by leaving it to decay for a short time before disposal as normal waste. Other isotopes used in medicine, with half-lives in parentheses, include:

It is a short synthetic peptide built from seven amino acids: methionine, glutamic acid, histidine, phenylalanine and three prolines. The sequence derives from the 4-10 fragment of adrenocorticotropic hormone with an added proline-glycine-proline tail. No plant or animal extract is involved; the material is produced by solid-phase peptide synthesis.
It was developed in the Soviet Union during the early 1980s by groups at the Institute of Molecular Genetics in Moscow. Russian approval for intranasal use followed, and it has been marketed there since. Western laboratories encountered it mainly through translated literature and, later, through online research-chemical trade.
It holds a Russian registration as a prescription nasal product but has no approval from the European Medicines Agency or the United States Food and Drug Administration. Buyers outside Russia usually receive material sold strictly for laboratory research. That distinction matters because research-grade and pharmaceutical-grade products carry different documentation and testing expectations.
It is described as a synthetic analogue of the ACTH(4–10) fragment, a short segment of adrenocorticotropic hormone. Its sequence differs from that fragment and includes two proline residues, which influence stability and behaviour in solution.
